1. What is MMP-3?
Matrix metalloproteinase-3 (MMP-3) is a useful marker to evaluate the prognosis of joint destruction in rheumatoid arthritis (RA), in Japan. MMP-3 is expressed in synovial tissues and involved in cartilage destruction in RA and osteoarthritis (OA). MMP-3 plays an important role in degradation and reconstitution of extracellular matrix by degrading proteoglycan, fibronectin, type IV collagen, laminin and type IX collagen. MMP-3 is secreted as a latent form and activated out of membrane, which participate in metabolism of matrix tissue. Activity of MMP-3 is inhibited specifically by tissue inhibitors of metalloprotienases (TIMPs).
2. Clinical Data in Japan
MMP-3 is produced in both synovial and cartilage tissues. MMP-3 is a substrate-specific enzyme for proteoglycans that consist of cartilages. So MMP-3 has been popular as one of the enzyme related for cartilage destruction in RA. 
Generally synovial membrane proliferates with inflammatory reaction along with progression of RA. It is thought that destruction of articular tissues in RA is caused by digestion by MMP-3, that increases following to synovial inflammation and proliferation.
In current study, it is suggested that following to synoviocyte proliferation in RA, MMP-3 is expressed and produced in surface of synovial cells, and enzyme activity of MMP-3 might destruct articular tissues. MMP-3 from synoviocyte accumulates in synovial fluid, moves to blood circulation through blood and lymphatic vessels, and then serum revel of MMP-3 might increase.
Based on those data in-vivo, it is indicated that monitoring of serum MMP-3 level may be useful as a marker of synovial proliferation in early RA, and as a prognostic marker of articular destruction in RA.

Serum MMP-3 revel reflects synovial MMP-3 levels, and it indicates inflamed synovial membrane quantitatively which will be destructed. In this data, it is suggested that
Serum MMP-3 level reflects the extent of synovial proliferations in RA.
Serum MMP-3 is already elevated in early RA in a year.

Those data suggest that Monitoring of serum MMP-3 level is useful as prognostic marker for predicting prognosis of articlar destruction in early RA.
In the monitoring of early RA, with evaluating bone destruction as “Larsen’s Score”, patients those serum MMP-3 levels indicated high or elevated at first diagnosis, after a year, and during monitoring after two to three years, are progressive RA. But patients those serum MMP-3 levels indicated low or declined are non-progressive RA.

4. Assay Principal
This premier “PANACLEAR MMP-3 「PLATE」 kit is based on a one-step sandwich enzyme immunoassay using two different mouse anti MMP-3 monoclonal antibodies and quantifies MMP-3 by a simple one-step ELISA method.
