        <![CDATA[<p>Alzheimer's disease (AD) was first reported by A. Alzheimer, a German neuropathologist in 1907 and is considered as a major factor of dementia. It is known that Amyloid &beta; (A&beta;; which is major constituent of senile plaque) is cleaved from Amyloid Precursor Protein (APP; which exists in three main isoforms, APP695, APP751, and APP770) by &beta;-secretase and subsequent &gamma;-secretase . The production of soluble APP&beta; (sAPP&beta;) by &beta;-secretase cleavage corresponds to A&beta; production accordingly, so it is desired to measure sAPP&beta; in parallel with A&beta;. <br />In addition, it is reported that APP gene mutation exists in early-onset familial Alzheimer's disease patient. Swedish mutation, one of the APP gene mutations, is a double mutation at positions -1 to -2 from the &beta;-secretase cleavage site (Lys670&rarr;Asn and Met671&rarr;Leu). And further, it is reported that Swedish mutation elevates A&beta;40 and A&beta;42 production , and that the mutation is utilized in establishment of transgenic mice . <br />The measuring sAPP&beta; in Swedish type is useful for research of AD as well as in wild type. On the one hand, it is considered that in the metabolic pathway of APP, APP is first cleaved by &alpha;-secretase rather than &beta;-secretase normally to produce soluble APP&alpha; (sAPP&alpha;) and subsequently P3 is cleaved from the remaining C-terminal fragment by &gamma;-secretase. In recent research, there are several attempts to apply the inhibitor of &beta;-secretase and the activation of &alpha;-secretase for AD treatment.</p>

<p><strong><span style="font-family: sans-serif;"><span style="font-size: small;"><br />Abstract:</span></span></strong></p>
<p><span style="font-family: sans-serif;"><span style="font-size: small;"><br />The correct identification of circulating molecular forms and measurement of peptide levels in blood entails that the endocrine peptide being studied is stable and recovered in good yields during blood processing. However, it is not clear whether this is achieved in studies using standard blood processing. Therefore, we compared peptide concentration and form of 12 (125)I-labeled peptides using the standard procedure (EDTA-blood on ice) and a new method employing Reduced temperatures, Acidification, Protease inhibition, Isotopic exogenous controls, and Dilution (RAPID). During standard processing there was at least 80% loss for calcitonin-gene-related peptide and cholecystokinin-58 (CCK-58) and more than 35% loss for amylin, insulin, peptide YY forms (PYY((1-36)) and PYY((3-36))), and somatostatin-28. In contrast, the RAPID method significantly improved the recovery for 11 of 12 peptides (P &lt; 0.05) and eliminated the breakdown of endocrine peptides occurring after standard processing as reflected in radically changed molecular forms for CCK-58, gastrin-releasing peptide, somatostatin-28, and ghrelin. For endogenous ghrelin, this led to an acyl/total ghrelin ratio of 1:5 instead of 1:19 by the standard method. These results show that the RAPID method enables accurate assessment of circulating gut peptide concentrations and forms such as CCK-58, acylated ghrelin, and somatostatin-28. Therefore, the RAPID method represents an efficacious means to detect circulating variations in peptide concentrations and form relevant to the understanding of physiological function of endocrine peptides.<br /></span></span></p>

Corticosterone can be used as a non-invasive biomarker of stress study through the collection of urine and feces to avoid corticosterone increase of blood levels which is caused by normal invasive methods.<br /></span></span></p>

        <![CDATA[<p><span style="font-size: small;">Peroxisome proliferator-activated receptor gamma (PPARg; NR1C3) is a member of orphan nuclear receptor. Oxidized metabolites of linoleic acid, 9- hydroxyctadienoic acid (9-HODE) and 13-HODE are activators and ligands of PPARg.<br />PPARg plays important roles in lipid and glucose metabolism, and have been implicated in obesity-related metabolic diseases such as hyperlipidemia, insulin resistance, and coronary artery disease. Three members were called PPARa, b, g. Three N-terminal isoforms, called g1, g2 and g3, are known to arise by alternative splicing and promoter usage from the PPARg gene. RXR is an obligate partner for PPAR.<br /><br />Those antibodied are very useful for analogue drug study, and obesity related diseases study.<br /></span></p>

As the improved kit can also measure mouse PYY, then the kit name is changed to YK081 Mouse/Rat PYY EIA Kit. The minor changes were done in the improved kit, please read the entire package insert carefully before beginning the assay.<br /><br /><br /><br /><br /></span></span></p>
